Näytelmä kertoo toimittaja Frances Thurstonista, jonka setä professori George Angell kuolee epämääräisissä olosuhteissa. Käydessään setänsä jäämistöä läpi Thurston törmää arkistolaatikkoon, jonka sisällä odottaa mysteeri: selontekoja, unitutkimuksia, päiväkirjoja, lehtileikkeitä ja kammottava savityö.

Cthulhun kutsu perustuu kulttikirjailija H.P. Lovecraftin (1890–1937) samannimiseen novelliin. Teos on Lovecraftin tunnetuimpia tarinoita ja tärkeä osa niin sanottua Cthulhu Mythosta, ”kauhujen pantheonia”, joka on toiminut innoittajana monille eri alojen taiteilijoille 1900-luvun alusta lähtien.
